The expressed class C-like β-lactamase gene, from Methanosarcina sp., also shows hydrolysis activity on nitrocefin and is more closely related to DD-peptidase enzymes. Our findings highlight the need to redefine the nomenclature of β-lactamase enzymes and the specification of multipotent enzymes in different ways in Archaea and bacteria over time.Immunotherapy targeting T-cell inhibitory receptors, namely programmed cell death-1 (PD-1) and/or cytotoxic T-lymphocyte associated protein-4 (CTLA-4), leads to durable responses in a proportion of patients with advanced metastatic melanoma. Combination immunotherapy results in higher rates of response compared to anti-PD-1 monotherapy, at the expense of higher toxicity. Currently, there are no robust molecular biomarkers for the selection of first-line immunotherapy. We used flow cytometry to profile pretreatment tumor biopsies from 36 melanoma patients treated with anti-PD-1 or combination (anti-PD-1 plus anti-CTLA-4) immunotherapy. A novel quantitative score was developed to determine the tumor cell expression of antigen-presenting MHC class I (MHC-I) molecules, and to correlate expression data with treatment response. Melanoma MHC-I expression was intact in all tumors derived from patients who demonstrated durable response to anti-PD-1 monotherapy. In contrast, melanoma MHC-I expression was low in 67% of tumors derived from patients with durable response to combination immunotherapy. Compared to MHC-I high tumors, MHC-I low tumors displayed reduced T-cell infiltration and a myeloid cell-enriched microenvironment. Our data emphasize the importance of robust MHC-I expression for anti-PD-1 monotherapy response and provide a rationale for the selection of combination immunotherapy as the first-line treatment in MHC-I low melanoma.For the last 30 years at the Royal Hospital for Women, unifocal vulvar squamous cancers have been treated by radical local excision, aiming to achieve a histopathological margin of ?8 mm, equating to a surgical margin of 1 cm. The need for a margin of this width has recently been challenged. We aimed to determine the long-term outcome following this conservative approach, and the relationship between vulvar recurrences and surgical margins. Data were obtained retrospectively on 345 patients treated primarily with surgery for squamous vulvar cancer between 1987 and 2017. Median follow-up was 93 months. Five-year disease-specific survival was 86%. Of 78 vulvar recurrences, 33 (42.3%) were at the primary site and 45 (57.7%) at a remote site. In multivariable analysis, a margin less then 5 mm showed a higher risk of all vulvar (Hazard ratio (HR), 2.29; CI, 1.12-4.70), and primary site recurrences (subdistribution hazard ratio (SHR), 15.20; CI, 5.21-44.26), while those with a margin of 5 to less then 8 mm had a higher risk of a primary site recurrence (SHR, 8.92; CI, 3.26-24.43), and a lower risk of remote site recurrence. Excision margins less then 8 mm treated by re-excision or radiation therapy had a significantly decreased risk of recurrence. Guidelines should continue to recommend a surgical margin of 1 cm.Using Automated Planning for the high level control of robotic architectures is becoming very popular thanks mainly to its capability to define the tasks to perform in a declarative way. However, classical planning tasks, even in its basic standard Planning Domain Definition Language (PDDL) format, are still very hard to formalize for non expert engineers when the use case to model is complex. Human Robot Interaction (HRI) is one of those complex environments. This manuscript describes the rationale followed to design a planning model able to control social autonomous robots interacting with humans. It is the result of the authors' experience in modeling use cases for Social Assistive Robotics (SAR) in two areas related to healthcare Comprehensive Geriatric Assessment (CGA) and non-contact rehabilitation therapies for patients with physical impairments. In this work a general definition of these two use cases in a unique planning domain is proposed, which favors the management and integration with the software robotic architecture, as well as the addition of new use cases. Results show that the model is able to capture all the relevant aspects of the Human-Robot interaction in those scenarios, allowing the robot to autonomously perform the tasks by using a standard planning-execution architecture.Cooperative transmission is a promising technology for underwater acoustic sensor networks (UASNs) to ensure the effective collection of underwater information. In this paper, we study the joint relay selection and power allocation problem to maximize the cumulative quality of information transmission in energy harvesting-powered UASNs (EH-UASNs). First, we formulate the process of cooperative transmission with joint strategy optimization as a Markov decision process model. In the proposed model, an effective state expression is presented to better reveal interactive relationship between learning and environment, thereby improving the learning ability. Then, we further propose a novel reward function which can guide nodes to adjust power strategy adaptively to balance instantaneous capacity and long-term quality of service (QoS) under the dynamic unpredictable energy harvesting. More specifically, we propose a deep Q-network-based resource allocation algorithm for EH-UASNs to solve the complex coupled strategy optimization problem without any prior underwater environment information. Finally, simulation results verify the superior performance of the proposed algorithm in improving the cumulative network capacity and reducing outages.In this study we report the preparation of novel multicomponent hydrogels as potential biomaterials for injectable hydrogels comprised of alginate, casein and bacterial cellulose impregnated with iron nanoparticles (BCF). These hydrogels demonstrated amide cross-linking of alginate-casein, ionic cross-linking of alginate and supramolecular interaction due to incorporation of BCF. The physico-chemical properties of gels were characterized via FTIR, XRD, DSC, TGA, VSM and mechanical compression. The results indicated that incorporation of BCF in alginate/casein hydrogels led to mechanically stronger gels with magnetic properties, increased porosity and hence increased swelling.
